Description
Background:
Streptavidin is a biotin binding protein present in the fermentation broth of the bacterium Streptomyces avidinii. Each molecule of streptavidin can bind four molecules of biotin with a high affinity constant (Kd~10 -15). Unlike native avidin, streptavidin is not glycosylated and has a near neutral isoelectric point (pI ~ 5-6) vs a pI of 10 for native avidin.

Alkaline Phosphatase (Alk Phos) is an enzyme purified from bovine calf intestinal mucosa with a molecular weight of approximately 140 kDa.

Streptavidin-Alk. Phos. is supplied in 0.01M Tris pH 8.0, containing .25 NaCl, 30 mg/ml BSA and 0.05% sodium azide as a preservative.

Storage and Stability
This Alkaline Phosphatase conjugate is stable for ~6 weeks when stored at 2-8°C. Do not freeze. For extended storage, add an equal volume of ACS grade or better glycerol for a final concentration of 50%, then store at -20°C.
Applications and Suggested Working Dilutions
Western blotting: 0.1 - 1.0 µg/ml
ELISA: 1 - 2 µg/ml
Immunohistochemistry: 1 - 2 µg/ml
Western blotting and ELISA Chemiluminescent: 0.1 - 0.05 µg/ml

However, each investigator should determine their own optimal working dilution for each specific research application.
