This monoclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 - 7.3 containing 5.0% w/v trehalose with no calcium, magnesium or preservatives present.
Storage and Handling
The lyophilized antibody can be stored desiccated at -20°C to -70°C for up to twelve months. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid repeated freeze thaw cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Human ALK-1 this monoclonal antibody can be used at a concentration of 1-2 µg/ml. This monoclonal antibody should be used in conjunction with compatible second-step reagents such as PN:M114 and a chromogenic substrate such as PN:T343. The detection limit for Human ALK-1 is 5.0 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is PN:A343.
Simple Western: Suitable for use at concentration of 40 µg/mL.

Specificity
Mouse Anti-Human Activin Receptor-Like Kinase 1 (ALK-1) (Clone 117720) recognizes an epitope on Human ALK-1. This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype. This antibody shows less than 1% cross-reactivity with rmALK-1, rhActivin RIA, and rhActivin RIB.

Background
Activin A receptor type II-like 1, also known as ACVRL1, is a human gene. ACVRL1 is a receptor in the TGF beta signaling pathway. It is also known as "activin receptor-like kinase 1", or "ALK1". This gene encodes a type I cell-surface receptor for the TGF-beta superfamily of ligands. It shares with other type I receptors a high degree of similarity in serine-threonine kinase subdomains, a glycine- and serine-rich region (called the GS domain) preceding the kinase domain, and a short C-terminal tail. The encoded protein, sometimes termed ALK1, shares similar domain structures with other closely related ALK or activin receptor-like kinase proteins that form a subfamily of receptor serine/threonine kinases. Mutations in this gene are associated with hemorrhagic telangiectasia type 2, also known as Rendu-Osler-Weber syndrome 2 suggesting a critical role for ALK-1 in the control of blood vessel development or repair.

Human and mouse ALK-1 share approximately 71% amino acid sequence identity in their extracellular regions.
