Description
Specificity:
Anti-Mouse CD11b (Mac-1) recognizes the (Mr 170 kD) αm subunit of Mac-1 (CD11b/CD18, αmβ2 integrin), the mouse macrophage-granulocyte specific antigen. Mac-1 is a macrophage differentiation antigen associated with type three complement receptor and mediates adhesion to CD54 (ICAM-1).1

Antigen Distribution:
The Mac-1 CD11b antigen is present on macrophages, granulocytes, natural killer cells, blood monocytes. CD11b is expressed on 8% spleen cells, 44% bone marrow cells and less than 1% of thymocytes.

Formulation
This purified antibody is formulated in 0.01 M phosphate buffered saline (PBS) pH 7.4, 150 mM NaCl, 1% BSA and 0.09% sodium azide as a preservative.
Storage and Stability
This antibody is stable when stored at 2-8°C. Do not freeze.
Reported Applications
IHC (Frozen), FC, IP, B
Immunohistochemistry on Frozen Sections: This antibody should give satisfactory staining results when used on frozen sections at a concentration of 2.5 µg/ml.
Flow Cytometry: It is recommended to use the indirect method for signal enhancement when enumerating cells expressing CD11b. A suggested method would be to stain cells expressing CD11b with 0.25 µg per 1 x 106 cells in a total staining volume of 100 µl followed by Goat F(ab')2 Anti-Rat IgG, Fc (Gamma) Fragment Specific-R-phycoerythrin (Leinco Prod. No.: R485).
Blocking: It is recommended to use Anti-Mouse CD11b (Mac-1 αm chain) Purified Low Endotoxin Functional Formulation (Leinco Prod. No.: C377) for functional studies
Each investigator should determine their own optimal working dilution for specific applications.
