Description
Specificity:
Mouse Anti-Human Beta-Nerve Growth Factor (β-NGF) (Clone 25623) recognizes an epitope on Human β-NGF. This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype. This antibody shows strong cross-reactivity with rhBDNF, rhNT-3 and rhNT-4. However, this antibody did not neutralize the biological activity of rhBDNF, rhNT-3 or rhNT-4. When immobilized on a microplate, this antibody will capture both recombinant as well as natural human β-NGF. When used in combination with the biotinylated anti-human β-NGF detection antibody (Catalog # BAF256) in sandwich ELISAs, less than 5% cross-reactivity was observed with rrβ-NGF, rmβ-NGF, rhBDNF, rhNT-3 or rhNT-4
Background:
Nerve growth factor beta (NGF-beta) is a secreted protein that is important for the differentiation and survival of neurons and dermal keratinocytes.1 It is also involved in cell proliferation and survival, and it is a mediator of the immune response.2 NGF-beta accelerate ulcer healing.3

Formulation
This monoclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 - 7.3 containing 5.0% w/v trehalose with no calcium, magnesium or preservatives present.
Reconstitution
This lyophilized antibody may be aseptically reconstituted with sterile PBS to a final volume of 1 ml. This will result in a final antibody concentration of 500 µg/ml.

<0.1 EU/µg as determined by the LAL method
Storage and Stability
The lyophilized antibody can be stored desiccated at -20°C to -70°C for up to twelve months. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid repeated freeze thaw cycles. No detectable loss of activity was observed after six months.

ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 2 µg/ml. A suitable detection antibody is Anti-Human β-NGF-Biotin (Antigen Aff. Pur.) (
Leinco Prod. No.: N127) at a concentration of approximately 0.1-0.4 µg/ml. A suggested standard for this assay is Recombinant Human β-NGF (
Leinco Prod. No.: N116).
Neutralization: This antibody is useful for neutralization of Human β-NGF bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Human β-NGF (at 5.0 ng/ml) is 0.1 - 0.4 µg/ml.
Each investigator should determine their own optimal working dilution for specific applications.
