Description
Specificity:
Goat Anti-Mouse Flt-3 Ligand recognizes Mouse Flt-3 Ligand. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.

Background:
Fms-related tyrosine kinase 3 ligand (FLT3LG) is a hematopoietic four helical bundle cytokine. It is structurally homologous to stem cell factor (SCF) and colony stimulating factor 1 (CSF-1). In synergy with other growth factors, such as IL-7 and IL-3, Flt-3 ligand promotes long-term expansion and differentiation of human pro-B-cells.1,2 Flt3 ligand is a cytokine capable of inducing large numbers of dendritic cells that dramatically enhances the sensitivity of antigen-specific B and T cell responses to systemic injection of a soluble protein. Through a CD40-CD40 ligand-dependent mechanism Flt-3 Ligand influences the class of antibody produced and enables productive immune responses.3

Formulation
This antigen affinity purified polyclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 containing 5.0% w/v trehalose with no calcium, magnesium, or preservatives present.
Reconstitution
We suggest aseptically reconstituting this lyophilized, antigen affinity purified polyclonal antibody with 1.0 ml of sterile 1X PBS pH 7.2-7.3. This will result in a final antibody concentration of 0.1 mg/ml.

<0.1 EU/µg as determined by the LAL method
Storage and Stability
The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

ELISA Cap, N, WB
ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 0.4 µg/ml. A suitable detection antibody is Anti-Mouse Flt-3 Ligand-Biotin (Antigen Aff. Pur.) (Leinco Prod. No.: F1096) at a concentration of approximately 200 ng/ml. When used in conjunction with an optimal second step reagent such as UltraAvidin™- Horseradish Peroxidase (HRPO) (Leinco Prod. No.: A106) and a chromogenic substrate such as TMB HRPO Microwell Substrate Standard Kinetic One Component "Ready Use" (Leinco Prod. No.: T118), the sensitivity of this sandwich ELISA is ~1.0 ng/well. A suggested standard for this assay is Recombinant Mouse Flt-3 Ligand (Leinco Prod. No.: F159).
Neutralization: This antibody is useful for neutralization of Mouse Flt-3 Ligand bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Mouse Flt-3 Ligand (at 10 ng/ml) is 0.05-0.25 µg/ml.
Western Blotting: To detect Mouse Flt-3 Ligand this polyclonal antibody can be used at a concentration of 0.1-0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as Donkey Anti-Goat IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: G505) and a chromogenic substrate such as TMB HRPO Membrane/IHC Substrate UltraSensitive™ One Component "Ready Use" (Leinco Prod. No.: T343). The detection limit for Mouse Flt-3 Ligand is 5 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is Recombinant Mouse Flt-3 Ligand (Leinco Prod. No.: F159).
Each investigator should determine their own optimal working dilution for specific applications.
