Description
Background:
Interleukin-2 (IL-2), also known as TCGF, is a secreted cytokine that is important for the proliferation of T and B lymphocytes (1-2). IL-2 also stimulates growth and differentiation of B cells, NK cells, LAK cells, monocytes, and oligodendrocytes (3). The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by IL-4 and IL-7. The expression of the IL-2 gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli. IL-2 has been shown to have antitumor effects in some studies which are probably mediated by cytotoxic effector cells (4).

Biological Activity
The biological activity of Human IL-2 was determined in a cell proliferation assay using an IL-2 dependent murine cytotoxic T cell line, CTLL-2 (Gearing, A.J.H. and C.B. Bird in Lymphokines and Interferons, a Practical Approach. 1987, M.J. Clemens, A.G. Morris and A.J.H. Gearing, eds., IRL Press, p. 295). The expected ED50 for this effect is typically 0.25 - 0.5 ng/ml.
Each investigator should determine their own optimal working dilution for specific applications.
