Description
Background:
The B-lymphocyte activation antigen B7-1, also known as CD80, is a member of cell surface immunoglobulin superfamily. It is expressed on the surface of antigen-presenting cells including activated B cells, macrophages and dendritic cells. As costimulatory ligands, B7-1 which exists predominantly as a dimer and the related protein B7-2, interact with the costimulatory receptors CD28 and CTLA-4 expressed on T cells, and thus constitute one of the dominant pathways that regulate T cell activation and tolerance, cytokine production and the generation of CTL (1). Binding of this protein with CTLA-4 is involved in down-regulation of immune response. Contacts between B and T helper cells mediated by B7-1 provide negative signal for the proliferation and IgG secretion of normal B cells and B cell lymphomas. In addition, B7-1 also plays a role in induction of innate immune responses by activating NF-κB signaling pathway in macrophages. B7-1 is thus regarded as promising therapeutic targets for autoimmune diseases and various carcinomas (3).

The predicted molecular weight of Recombinant Human B7-1 is Mr 51.3 kDa (monomer). However, the actual molecular weight as observed by migration on SDS Page is Mr 70-90 kDa.

Formulation
This recombinant protein was 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 with no calcium, magnesium, or preservatives.
Purity
>95% by SDS-PAGE and analyzed by silver stain.
Storage and Stability
This lyophilized protein is stable for six to twelve months when stored desiccated at -20°C to -70°C. After aseptic reconstitution, this protein may be stored at 2°C to 8°C for one month or at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. Biological Activity
The biological activity of Human B7-1 was determined by its ability to induce IL2 secretion by Jurkat human leukemic T cells. The expected ED50 is 0.3-0.9 μg/ml in the presence of PHA.
Each investigator should determine their own optimal working dilution for specific applications.
