Background

Fibroblast growth factor 6, also known as FGF6, hst-2 or hst-1-related is a member of the fibroblast growth factor (FGF) family. 1,2 FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. FGF6 is the only known member of the FGF family whose expression is restricted to the muscle cell lineage during development, suggesting it may have a role in myogenesis.3 FGF6 has a dual function in muscle regeneration, stimulating myoblast proliferation/migration and muscle differentiation/hypertrophy.4

Biological Activity
The biological activity of Human FGF-6 was monitored in a mitogenic assay by measuring the FGF dependent 3H-thymidine incorporation in quiescent NR6R-3T3 fibroblasts (Thomas, K. 1987, Methods in Enzymology 147:120-135). The expected ED<sub>50</sub> for this effect is typically 0.1 - 0.3 ng/mL in the presence of 1 μg/mL of heparin.
Amino Acid Sequence
The DNA sequence encoding the putative 168 amino acid mature FGF-6 was cloned into a suitable expression vector and expressed in E. coli (Coulier, F. et al., 1991, Oncogene 6:1437; Iida, S. et al ., 1992, Oncogene 7:303).
