The lyophilized, biotinylated antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for up to twelve months from date of receipt. The reconstituted biotin conjugate can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted conjugate, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Human IL-3 this biotin conjugate can be used at a concentration of 0.1 - 0.2 µg/ml. This biotin conjugate should be used in conjunction with compatible second-step reagents such as PN:A106 and a chromogenic substrate such as PN:T343. The detection limit for Human IL-3 is 2 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is PN:I-185.
ELISA Sandwich Assay: This antibody can be used as the detection antibody in a sandwich ELISA at a concentration of approximately 0.1-0.4 µg/ml when used in conjunction with PN:I-686 as the capture antibody at 2-8 µg/ml and an optimal second step reagent such as PN:A106 for the detection of Human IL-3.

This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.
Background
IL-3 is a 17 kD pleiotropic cytokine and a member of a family of growth factors that sustains both the proliferation and the development of hematopoietic precursors. It also displays neurotrophic activity and is thought to be associated with neurologic disorders. Signal transmission is assured by high affinity binding to the IL-3 receptor (IL3RA). The receptor contains α and β subunits. IL-3 shares the β subunit with IL-5 and GM-CSF which explains their biological functional similarities. The IL-3/receptor complex induces JAK2/STAT5 cell signalization pathway and it can stimulate both the activation of gene expression and the suppression of apoptosis via transcription factor c‑myc and the Ras pathway, respectively. IL-3 fuels the differentiation of multipotent hematopoietic stem cells into myeloid progenitor cells or lymphoid progenitor cells (when in conjunction with IL-7). Together with other cytokines, including EPO, GM-CSF, and IL-6, IL-3 can stimulate proliferation of all cells in the myeloid lineage. The constitutive expression of IL-3 in some leukemia cell lines (as opposed to selective T-cell expression in response to specific impulses or antigens) is thought to fuel the development of the disease. Post-chemotherapy IL-3 application can promote regeneration of granulocytes and platelets. Furthermore, during high-dose chemotherapy, the combination of IL-3, GM-CSF and stem cell factor increases peripheral blood stem cells. IL-3 is thought to have therapeutic potential in lymphohematopoietic disorders and solid cancers.
