Background:
Sonic hedgehog (SHH) is a secreted factor regulating patterning of the anterior-posterior axis in the developing limb.1 SSH functions as a secreted signaling molecule in limb pattern formation, differentiation of motor neurons, and sclerotome induction.2 SHH pathway is known to be functional in the developing male tract.3 SHH may have therapeutic value as a protective agent in neurodegenerative disease.4

This recombinant protein was 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 with 0.3 M NaCl, no calcium, magnesium, or preservatives.
Purity
>95% by SDS Page and analyzed by silver stain.
Storage and Stability
This lyophilized protein is stable for six to twelve months when stored desiccated at -20°C to -70°C. After aseptic reconstitution, this protein may be stored at 2°C to 8°C for one month or at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. Biological Activity
The biological activity of recombinant Human Sonic Hedgehog is determined by its ability to induce alkaline phosphatase production by C3H/10T1/2 (CCL-226) cells. The expected ED50 is 0.1-0.4 μg/ml.
Each investigator should determine their own optimal working dilution for specific applications.
Amino Acid Sequence
The DNA sequence encoding amino acid residues Cys 24 - Gly 197 of human SHH with Ile-Ile substituted for Cys 24 (Riddle, R.D. et al., 1993, Cell 75:1401 - 1416; Taylor, F.R. et al., 2001, Biochemistry 40:4359 - 4371) was inserted into a suitable expression vector and expressed in E. coli.
