Description
Specificity:
Anti-Human Cytokeratin 19 (clone BA17) reacts with the smallest human cytokeratin (Mr 40 kDa).1 The antibody reacts specifically with cytokeratin 19 and has no reactivity with other cytokeratins.

Antigen Distribution:
Anti-Cytokeratin 19 reacts strongly with luminal (secretory) epithelial cells but does not stain the basal (myoepithelial) cells. There is a subset of secretory epithelial cells which were Keratin 19- and are thought to be precursors to keratin 19+ cells.1,2

Background:
Cytokeratin 19, a type I intermediate filament protein, it is the smallest known acidic cytokeratin. The degree of keratin 19 positivity in breast cancer distinguishes malignant from benign tumours.

Formulation
This purified antibody is formulated in 0.01 M phosphate buffered saline (PBS) pH 7.4, 150 mM NaCl, 1% BSA and 0.09% sodium azide as a preservative.
Storage and Stability
This antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at – 20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

IHC (NBF/Par.), WB, ICC
IHC (NBF/Par.): This Antibody has not been tested by Leinco for the use in immunohistochemistry. However, the antibody was used in reference 1 for immunohistochemistry.
Western Blotting: To detect Human KRT19 this monoclonal antibody can be used at a concentration of 0.1-0.2 µg/ml. This monoclonal antibody should be used in conjunction with compatible second-step reagents such as Goat Anti-Mouse IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: M114) and a chromogenic substrate such as TMB HRPO Membrane/IHC Substrate UltraSensitive™ One Component "Ready Use" (Leinco Prod. No.: T343). The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.
Immunocytochemistry: This antibody was used at a concentration of 10 μg/mL to detect Cytokeratin 19 on human MDA-MB 231 cells. Cells were fixed with PBS containing 4% paraformaldehyde for 45 minutes at room temperature and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA for 1 hour at room temperature. After blocking, cells were incubated with diluted primary antibody overnight at 4° C followed by Rhodamine Red-coupled anti-mouse IgG at room temperature covered with foil for one hour. Between each step, cells were washed with Leinco Prod. No. W101.
Each investigator should determine their own optimal working dilution for specific applications.
