State of Matter Liquid Components Serion RF Absorbent - 20 mL (100 Tests) Country of Origin Germany DescriptionBackground SERION Rheumatoid Factor (Rf)-Absorbent for IgM-Antibody-Detection - Anti- human- IgG for the removal of IgM- rheumatoid factors from serum- and plasmaprobes or cerebrospinal fluid (CSF) which are to be prepared for the detection of specific IgM-antibodies in indirect Immunoassays. Principle of Method Rheumatoid factors are autoantibodies (anti-IgG) mainly of the IgM-class. They can be detected in the serum of approx. 5 % of healthy individuals. IgM-rheumatoid factors preferably bind to IgG-immune-complexes; therefore, false positive results may appear in the pathogen-specific IgM-detection. Furthermore, the possibility exists, that weakbinding pathogen-specific IgM antibodies are displaced by stronger-binding IgG antibodies. In this case, IgM-detection can bring about false negative results. In IgAdetection possibly occuring competition with IgG-antibodies can also be prevented by eliminating IgG with Rf-absorbent. The SERION Rf-absorbent precipitates up to 15 mg IgG/ml (undiluted serum). The IgMrheumatoid factor binds to the precipitate. Consequently, the IgG-antibodies as well as the IgM-rheumatoid factors are not reactive in the immunoassay. The use of SERION rheumatoid factor-absorbent can minimize false positive as well as false negative results. For Sale In the United States (USA) Only. Add 200 µl Rf-absorbent and 800 µl ELISA dilution buffer to 10 µl of the probe (yields serum dilution of 1+100). Mix well! 2. Incubate 15 minutes at room temperature or overnight at +2 to +8 C. Mix again. 3. Use 100 µl of the mixture for one test performance in the SERION ELISA classic. Test performance (CSF) 1. Predilution: Dilute Rf-absorbent with ELISA dilution buffer 1:5 (e.g. 100 µl + 400 µl) 2. Dilute CSF 1:2 or 1:10 with predilution (e.g. 100 µl + 100 µl or 20 µl + 180 µl). Mix well! Incubate 15 minutes at room temperature or overnight at +2 to +8°C. Mix again. 3. Use 100 µl of the mixture for one test performance in the SERION ELISA classic. Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change.
