2. Partial Delipidation Buffer (Component A) (B672) Kit contains 5mL 3. Partial Delipidation Buffer (Component B) (B676) Kit contains 5mL Note: Component B is corrosive to most plastic types; use glass 4. 1X ADAPT-3D Immunostaining Buffer (B673) Kit contains 15mL 5. Refractive Index Matching (Clearing Solution) (B675) Kit contains 15mL 6. Wash Buffer (5X) (B674) Kit contains 20mL Please note that required buffers and materials not included in the kit are: • Organoids • Positive and negative controls as required • Glass scintillation vials if partial delipidation is performed • Glass bottom 96-well plate • Primary and secondary antibodies • PBS pH 7.4 to dilute the 5x Wash Buffer and prepare the 50% RI equilibration solution • Fixative Solution – 4% paraformaldehyde (PFA) in PBS with 10% sucrose at pH 9.0 Storage and Handling Store all components at room temperature unless otherwise specified. The 1X ADAPT-3D™ Immunostaining Buffer (B673), which contains bovine serum albumin (BSA) and normal serum, must be stored refrigerated at 2–8 °C to ensure stability and prevent degradation. Do not freeze. Country of Origin USA Shipping Ambient Quality Testing and Recommended Usage The ADAPT-3D Organoid Clearing Kit from Leinco Technologies is meticulously manufactured under stringent quality standards, ensuring reliable and reproducible results for your 3D fluorescence imaging needs. Produced in our state-of-the-art facility in Saint Louis, Missouri USA, the ADAPT-3D Imaging Kit adheres to ISO 9001:2015 manufacturing standards. This commitment to excellence guarantees that every component of the ADAPT-3D protocol meets the highest benchmarks for consistency and performance. DescriptionBackground ADAPT-3D (Accelerated Deep Adaptable Processing of Tissue for 3-Dimensional Imaging) is designed to prepare organoids for 3D fluorescence imaging. Developed to leverage recent advances in aqueous-based clearing methods, ADAPT-3D allows for increased light penetration in organoid samples while simultaneously simplifying organoid preparation. This kit is specifically tailored for organoids grown in a 96 well glass bottom plate allowing for organoid fixation and imaging to occur on the same surface.
Benefits • Clears in minutes to a few hours. A 100–500 µm organoid reaches full transparency in the 100% RI matching step within ≈ 30 min to a few hours — not the days-to-weeks timescale of solvent, hydrogel, and hyperhydration protocols.
• No measurable change in organoid dimensions. Gentle partial delipidation replaces aggressive lipid stripping, dehydration, and hydrogel embedding, so volume, inter-cellular spacing, and network geometry survive processing intact. • Maximum fluorescence retention. The pH 9.0 fixative with 10% sucrose preserves sensitive endogenous reporters (eGFP, eYFP, tdTomato, mScarlet) without signal-boosting additives. Benchmarking returned ≈ 10× brighter antibody-conjugate signal and ≈ 3× brighter endogenous-reporter signal than a leading aqueous hyperhydration method. • One buffer to block, permeabilize, and detect. ADAPT-3D Immunostaining Buffer replaces the separate blocking, permeabilization, and antibody-dilution solutions of conventional whole-mount protocols. • Improved Workflows for Controls and Antibody Titrations. A full titration series, secondary-only wells, and positive and negative controls can be run in dedicated columns of the same plate, processed identically, and imaged on the same instrument. • Aqueous and non-hazardous. No regulated organic solvents, no specialized apparatus, and no microscope-objective incompatibility. • Image in place. Transfer the glass-bottom or optically clear-bottom plate directly to the microscope stage; well-to-well registration is preserved and mounting-induced distortion is eliminated. Directions for Use Please read the ADAPT-3D Organoid Clearing Product Insert for more information and detailed protocols for using this kit. Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change. Hazard Description The ADAPT-3D™ Imaging Kit contains multiple chemical components that may pose health and safety risks if not handled properly. These may include corrosive, irritant, or flammable substances. Some reagents can cause skin, eye, or respiratory irritation and may be harmful if inhaled or ingested. Certain components, such as the Partial Delipidation Buffer B, are particularly corrosive to plastics and require use in glass containers only. Always wear appropriate personal protective equipment, including gloves, lab coat, and safety glasses, and work in a well-ventilated area or fume hood.
For complete hazard classifications, precautionary statements, and emergency procedures, please consult the Safety Data Sheets (SDS) for each component before use.
