BackgroundAmiloride binding protein (ABP1), also referred to as diamine oxidase (DAO) or AOC1, is a ~110 kDa member of the copper-containing amine oxidase family. Naturally occurring as a glycosylated, disulfide-linked homodimer, AOC1 is primarily sequestered within the cytoplasmic granules of epithelial tissues in the kidney, placenta, uterus, lungs, and intestines. The enzyme is released into the extracellular environment upon induction by heparin. Functionally, AOC1 catalyzes the oxidative deamination of histamine and putrescine, yielding hydrogen peroxide and ammonia. This activity is critical for the catabolism of bioactive polyamines; consequently, AOC1 deficiencies are clinically linked to histaminosis and dietary histamine intolerance.
