DescriptionBackground Bovine enteric colibacillosis is an economically important disease in neonatal calves caused by enterotoxigenic E. coli (ETEC) which adhere to intestinal cells using pili. Most ETEC isolated from cattle use K99 pili to bind to enterocytes of the small intestine. K99 pili are primarily composed of a repeating 18.2kDa major subunit which is responsible for binding to host glycoprotein receptor, N-glycolylneuraminic acid-GM3. A 7.1kb operon of eight genes on a plasmid encode the pili and other associated proteins responsible for regulation, transport, and assembly of K99 pili. Background for Assay DevelopmentRapid and accurate diagnosis of Bovine enteric colibacillosis—a severe diarrheal disease in neonatal calves caused by enterotoxigenic E. coli (ETEC)—is critical for veterinary herd management. Because ETEC rely on K99 pili to adhere to the intestinal epithelium, the 18.2kDa major subunit of the K99 pilus serves as a highly reliable biomarker for diagnostic screening. The Leinco Technologies Anti-E. coli K99 Pili Antibody Library Pack is specifically engineered for In Vitro Diagnostic (IVD) manufacturers and assay developers. By providing a curated panel of four distinct, highly purified mouse monoclonal clones, this library pack enables rapid empirical screening. Assay developers can efficiently identify optimal matched pairs for capture and detection in high-sensitivity sandwich ELISAs, lateral flow assays (LFAs), and point-of-care veterinary diagnostic platforms. Key Advantages for IVD Manufacturers: 1.) Accelerated Matched Pair Screening: Evaluate four distinct Mouse IgG1 clones (K9911-201.3, K9912-349.2, K9912-418.2, and K9912-444.5) simultaneously to discover the highest-avidity combinations for your assay architecture. 2.) Exceptional Specificity: All included clones are rigorously validated to recognize K99+ E. coli. Crucially for diagnostic accuracy, they exhibit zero cross-reactivity to K99-negative strains (B41), E. coli K88ac, 987P, or other interfering veterinary pathogens. 3.) Seamless Scale-Up: Once you identify your optimal capture and detection clones from this library pack, Leinco provides consistent, reproducible bulk manufacturing and conjugation services to support commercial diagnostic kit production. Immunogen Purified E. coli K99 (B44) pili. Specificity These antibodies recognize K99+ E. coli. They do not cross-react with K99 negative E. coli (B41), E. coli K88ac, E. coli 987P, Proteus mirabilis, Proteus vulgaris, Pseudomonas aeruginosa, Enterobacter aerogenes, Citrobacter freundii, Klebsiella pneumoniae, or Strep. agalactiae. Product Preparation To support the stringent requirements of diagnostic assay developers and commercial kit manufacturers, this monoclonal antibody is produced in full compliance with dual ISO 13485 and ISO 9001 quality standards at our facility located in St. Louis, Missouri USA. The production workflow features a highly controlled, multi-step downstream purification sequence using affinity-grade Protein A or G. This method is optimized to deliver an ultra-pure formulation, maintaining critically low thresholds for bacterial endotoxins, leachable protein A, and high-molecular-weight aggregates.
