This recombinant monoclonal antibody is aseptically packaged and formulated in 0.01 M phosphate buffered saline (150 mM NaCl) PBS pH 7.2 - 7.4 with no carrier protein, potassium, calcium or preservatives added. Due to inherent biochemical properties of antibodies, certain products may be prone to precipitation over time. Precipitation may be removed by aseptic centrifugation and/or filtration.

Recombinant antibodies are manufactured in an animal free facility using only in vitro protein free cell culture techniques and are purified by a multi-step process including the use of protein A or G to assure extremely low levels of endotoxins, leachable protein A or aggregates.

This antibody may be stored sterile as received at 2-8°C for up to one month. For longer term storage, aseptically aliquot in working volumes without diluting and store at ≤ -70°C. Avoid Repeated Freeze Thaw Cycles.

Background
LIME1 is a lipid raft-associated transmembrane adaptor protein1. Lipid rafts play an essential role in the initiation of TCR, BCR, and FcR immunoreceptor signaling. In T cells, LIME1 is a positive regulator of TCR-mediated signaling pathways and plays roles in T cell development, activation, and differentiation2. Upon TCR stimulation, Lck phosphorylates LIME1, which then recruits various cytoplasmic proteins to membrane-proximal sites. LIME1 phosphorylation and association with intracellular signaling and effector molecules is controlled by CD4 and CD8 coreceptors1. In B cells, LIME1 augments BCR-mediated signaling after phosphorylation by Lyn, leading to NFAT and NF-κB pathway activation as well as B cell activation3.

EB0918-8H8-H2/K2 was generated in rabbit for use in immuno-MRM studies using a synthetic peptide derived from human LIME1 that is carbamidomethylated at the cysteines and phosphorylated at the threonine (pT274)4. However, the antibody exhibits low signal and fails to produce adequate linearity, and therefore its development for immuno-MRM assay use was halted. Additionally, EB0918-8H8-H2/K2 does not detect LIME1 by immunohistochemistry or Western blotting5.
Antigen Distribution
LIME1 is preferentially expressed by hemopoietic cells, particularly T cells and plasma cells, as well as in B cells, peripheral blood lymphocytes, and at low levels in thymocytes. LIME1 is found in lipid rafts and is recruited to the immunological synapse when T cells conjugate with antigen-presenting cells. LIME1 is also expressed in myeloma and plasmacytoma.
