DescriptionDescriptionSpecificity Nt 2535-2835 activity is directed against human and mouse ROR2, recognizing an epitope present in the ST2 domain. Nt 2535-2835 does not bind ROR1. Background ROR2 is a membrane-bound receptor tyrosine kinase activated by non-canonical Wnt signaling via its association with Wnt5A 1,2. ROR2 plays an essential role during normal bone and cartilage development 1 and is required to mediate cell migration during palate development in mammals 2. Wnt5a interaction with ROR2 inhibits Wnt/β-catenin signaling 1, which affects hair follicle formation, ossification in the calvarial mesenchyme, and mediates induction of osteoclastogenesis 3.
Dysregulation of ROR2 causes the diseases brachydactyly type B and Robinow syndrome, contributes to tumor progression in soft-tissue sarcomas, gastrointestinal stromal tumors 1, multiple myeloma, breast cancer, and cervical cancer 4, and has pro-tumorigenic effects in osteosarcoma, melanoma, and renal cell carcinoma cell lines 2. Additionally, upregulation of ROR2 is observed in osteoarthritis in humans, and blocking ROR2 improves cartilage integrity in osteoarthritis models by inhibiting yes-associated protein signaling 5. Nt 2535-2835 was generated by immunizing mice with the last 100 amino acids (bases 2535-2835) of the mouse ROR2 protein fused in frame to maltose binding protein 1. Splenic cells were fused and the resulting hybridomas screened via ELISA. Nt 2535-2835 is able to detect ROR2 in paraffin-embedded but not frozen tissues. Antigen Distribution ROR2 is expressed at the plasma membrane during embryonic development, predominantly in cartilage structures, and also in the growth plate, dermis, embryonic telencephalon, colon epithelium, and CD5+ B cells.
