State of Matter Lyophilized Product Preparation Purified by Protein A affinity chromatography Storage and Handling This product is stable for at least one (1) year at -20°C to -70°C. Reconstituted product should be stored in appropriate aliquots to avoid repeated freeze-thaw cycles. Country of Origin USA Shipping Next Day 2-8°C Applications and Recommended Usage? Quality Tested by Leinco ELISA: use at 0.1-1.0ug/ml
Immunoblotting: User should use at 1-5ug/ml. A band of ~60kDa is detected Immunocytochemistry: use at 2- 10ug/ml on virus-infected cells. These are recommended concentrations. End user should determine optimal concentrations for their applications. Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change. DescriptionSpecificity Mouse Monoclonal Antibody specific to Measles Nucleoprotein Background Measles virus (MeV) is an enveloped RNA virus in the Morbillivirus genus of the Paramyxoviridae family. Its non- segmented, negative-sense, single- stranded RNA genome is encapsidated by the viral nucleoprotein (N) within a helical nucleocapsid. Transcription and replication are carried out on this N:RNA complex by the viral polymerase complex which consists of two components, the large protein (L) and the phosphoprotein (P). MeV N consists of two regions: a structured N-terminal moiety, NCORE (aa 1–400), which contains all the regions necessary for self-assembly and RNA-binding, and a C-terminal domain, NTAIL(aa 401–525) that is intrinsically unstructured and is exposed at the surface of the viral nucleocapsid. Antigen DetailsFunction Encapsidates the genome in a ratio of 1 N per 6 ribonucleotides, protecting it from nucleases. The nucleocapsid (NC) has a helical structure with either 12.35 or 11.64 N per turn, approximately 20 nm in diameter, with a hollow central cavity approximately 5 nm in diameter. The encapsidated genomic RNA is termed the NC and serves as template for transcription and replication. During replication, encapsidation by N is coupled to RNA synthesis and all replicative products are resistant to nucleases. N is released in the blood following lysis of measles infected cells, it interacts then with human FCGR2B on immune cells, inducing apoptosis and blocking inflammatory immune response. Ntail binds to a protein on human thymic epithelial cells, termed Nucleoprotein Receptor (NR), inducing growth arrest.
