Specificity
M18/2 activity is directed against mouse CD18.
Background
Integrins are a large family of α/β heterodimeric transmembrane molecules that mediate adhesion, migration, cell survival, and cell differentiation1. The leukocytic cell-adhesion molecule (Leu-CAM) family has three members, LFA-1 (CD18/CD11a), CR3 (also known as MAC-1) (CD18/CD11b), and p150,95 (CD18/CD11c), all of which share the β chain CD182. Depending on which molecule CD18 is bound to, this β chain plays important roles in inflammation, initiation of the immune response, phagocytosis, adherence to endothelium, chemotaxis, and can act as a fibrinogen receptor as part of its heterodimer3.

M18/2 was generated by immunizing rat with mouse cytotoxic T cell glycoproteins and fusing the spleen cells with mouse myeloma line P3X63Ag8.6534. M18/2 was selected by immunoprecipitation of LFA-1 and found to bind to the β chain of both LFA-1 and MAC-1. M18/2 has subsequently been used to study lymphoma metastasis3,5, myelin phagocytosis6, platelet caspases and thrombocytosis7, and the immunosuppression of corneal allografts8.
