Specificity
34-1-2S activity is directed against H-2Dd and H-2Kd MHC Class I antigens. 34-1-2S cross-reacts with Kb, s, r, q, and p. Binding to H-2Kb is weak. 34-1-2S does not cross-react with H-2k and H-2f.
Background
H-2, the murine major histocompatibility complex (MHC), is composed of a diverse group of antigens divided into class I and II proteins that function in immune response1. MHC class I molecules bind peptides generated by the degradation of cytosolic proteins and then display those peptides on the cell surface. Generally, these peptides are derived from normal metabolism, but they can also be derived from foreign proteins during viral infection or allotransplantation. When peptides are recognized as foreign, cytotoxic T lymphocytes specific to the MHC class I- peptide complex kill the presenting cell.

34-1-2S was generated by immunizing C3H mice with spleen cells from (B6 X DBA/2) F1 mice2. Spleen cells were fused with SP2/0.Ag14 of BALB/c origin. 34-1-2S is cytotoxic to strains of many haplotypes. Deglycosylated 34-1-2S, and its Fab fragments, have abrogated binding to murine FcγRs compared with untreated or full-length antibodies3.

34-1-2S is known to induce transfusion-related acute lung injury and is used to study its pathogenesis3,4,5,6,7. Injection of 34-1-2S into BALB/c and other H-2d haplotype mice causes an H-2d–specific change in breathing accompanied by decreased blood oxygenation, increased lung water content, protein leakage into airways, and redistribution of blood neutrophils to the lungs6.
