CPTC Clone ID CPTC-Fas-1 Background Fas (also known as TNFRSF6, CD95) is a member of the tumor necrosis factor (TNF)
superfamily that induces apoptosis through activation of caspases1. When bound to its cognate
cytokine ligand (FasL), Fas triggers apoptosis or other forms of cell death via a six alpha helical
fold called a death domain. During this process, Fas recruits FADD, a receptor protein that
mediates recruitment of caspase-8, which then activates apoptosis via cleavage of downstream
substrates. Fas is also involved in the immune system1. Fas can potentiate a response to pathogens by activating NF-κB and promoting IL-1β production. Additionally, Fas co-stimulation during T cell activation can co-stimulate T cell proliferation and differentiation into short-lived effector memory T cells. Defects in Fas signaling can lead to autoimmunity (underexpression) or cancer (overexpression). Fas blockade is being investigated for the treatment of cancer. EB0111P-3F2-H2/K4 was generated in rabbit for use in immuno-MRM assays using a synthetic peptide derived from residues 289- 296 of human Fas that is phosphorylated at position Y291, EA(pY)DTLIK2. EB0111P-3F2-H2/K4 does not detect Fas by immunohistochemistry, reverse phase protein array, or Western blotting3.
