Antibody DetailsProduct DetailsReactive Species Human/Mouse/Rat Host Species Mouse Immunogen Fusion protein corresponding to aa 780-914 of the NR3A extracellular III-IV loop Product Concentration Lot Specific Formulation This monoclonal antibody is formulated in phosphate buffered saline (PBS) pH 7.2 - 7.4 with no carrier protein or preservatives added. State of Matter Liquid Product Preparation Antibodies are purified by a multi-step process including the use of protein A or G to assure extremely low levels of endotoxins, leachable protein A or aggregates. Storage and Handling Upon initial thawing, appropriately aliquot and store at -80°C. For long-term storage, keep at -80°C. Avoid repeated freeze-thaw cycles. Country of Origin USA Shipping Next Day 2-8°C Applications and Recommended Usage? Quality Tested by Leinco Immunoblotting: use at 1-10ug/mL. Predicted molecular weight 125-130kDa.
Immunohistochemistry: use at 1-10ug/mL. These are recommended concentrations. Endusers should determine optimal concentrations for their applications. Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change. DescriptionDescriptionSpecificity This antibody recognizes NMDAR subunits NR3A and NR3B. It does not cross-react with subunits NR1 or NR2A-D. Background Two subunits of the NMDA subtype of glutamate receptor (NMDAR) have been identified as NR3A and NR3B. These subunits show high sequence homology although different distribution patterns: NR3B is expressed predominantly in motor neurons while NR3A is more widely distributed. When expressed in Xenopus oocytes, NR3A or NR3B assemble with NR1 to form excitatory glycine receptors that are unaffected by glutamate. Function NMDA receptor subtype of glutamate-gated ion channels with reduced single-channel conductance, low calcium permeability and low voltage-dependent sensitivity to magnesium. Mediated by glycine. During the development of neural circuits, plays a role in the synaptic refinement period, restricting spine maturation and growth. By competing with GIT1 interaction with ARHGEF7/beta-PIX, may reduce GIT1/ARHGEF7-regulated local activation of RAC1, hence affecting signaling and limiting the maturation and growth of inactive synapses. May also play a role in PPP2CB-NMDAR mediated signaling mechanism. {UniProtKB:Q9R1M7}.
