Background
VTCN1 (also known as B7-H4) is a coinhibitory molecule that negatively regulates the activation and proliferation of T cells, plays an important role in the proliferation, invasion, and metastasis of tumors, and is protective against type 1 diabetes and in organ transplantation1. VTCN1 is a member of the B7/CD28 family which is generally involved in the activation of T cells and promoting immune tolerance. Translation of VTCN1 is strictly controlled in normal tissues, where VTCN1 RNA, but not protein, is widely detectable. However, VTCN1 protein is abnormally present in tumors, inflammation, and autoimmune diseases and is therefore a target of related immunotherapies. Several functional VTCN1 polymorphisms linked to increased risk of cancer have been identified. Additionally, increased VTCN1 protein abundance in tumor- associated macrophages is associated with shorter life expectancy2.

FSAI171R-7D11 was generated in mouse using a non-modified peptide sequence of human VTCN1 (EGVLGLVHEFK)3,4. FSAI171R-7D11 was developed for use in immuno-MRM assays and recognizes the EGVLGLVHEFK peptide as well as full-length recombinant VTCN1. The antibody does not work for detection in immunohistochemistry, immunofluorescence, or Reverse Phase Protein Array5. FSAI171R-7D11 can detect VTCN1 recombinant protein by Western blotting but does not detect VTCN1 in tissue or cell lysate samples.
Antigen Distribution
VTCN1 RNA, but not protein, is widely expressed in most normal tissues. VTCN1 protein is detected in ovarian, breast, renal cell, endometrial, and non-small-cell lung cancers as well as immunosuppressive tumor-associated macrophages. VTCN1 is present on cell membrane surfaces, but a soluble form can also be found.
