The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Human CCL14 this polyclonal antibody can be used at a concentration of 0.1 - 0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as PN:G505 and a chromogenic substrate such as PN:T343. The detection limit for Human CCL14 is 1 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is H257.

Neutralization: Measured by its ability to neutralize CCL14a/HCC‑1-induced chemotaxis in the BaF3 mouse pro‑B cell line transfected with human CCR1. The Neutralization Dose (ND50) is typically 0.15-0.75 µg/mL in the presence of 10 ng/mL Recombinant Human CCL14a/HCC‑1 aa 28‑93.

Background
Chemokine (C-C motif) ligand 14 (CCL14), also known as HCC-1, is a small cytokine belonging to the CC chemokine family. It is expressed in various tissues including spleen, bone marrow, liver, muscle, and gut.1 CCL13 activates monocytes, but does not induce their chemotaxis. CCL14 is of recent interest because of its natural ability to bind to and signal through the human immunodeficiency virus type-1 (HIV-1) co-receptor, CC chemokine receptor 5 (CCR5).2
