This recombinant monoclonal antibody is aseptically packaged and formulated in 0.01 M phosphate buffered saline (150 mM NaCl) PBS pH 7.2 - 7.4 with no carrier protein, potassium, calcium or preservatives added. Due to inherent biochemical properties of antibodies, certain products may be prone to precipitation over time. Precipitation may be removed by aseptic centrifugation and/or filtration.

Recombinant antibodies are manufactured in an animal free facility using only in vitro protein free cell culture techniques and are purified by a multi-step process including the use of protein A or G to assure extremely low levels of endotoxins, leachable protein A or aggregates.

This antibody may be stored sterile as received at 2-8°C for up to one month. For longer term storage, aseptically aliquot in working volumes without diluting and store at ≤ -70°C. Avoid Repeated Freeze Thaw Cycles.

CPTC-CD40-1

CD40 (also known as TNFRSF5) and CD40L (TNFSF5) are a receptor ligand pair belonging to the tumor necrosis factor receptor/TNF superfamily that functions in immune regulation¹. CD40 acts as a co-stimulatory receptor that, when interacting with its ligand CD40L on activated T cells, promotes activation, survival, production of pro-inflammatory cytokines and chemokines in dendritic cells, macrophages, and B cells, and enhances tumor antigen presentation^1,2. This in turn can further activate tumor-infiltrating lymphocytes². CD40 is a target of immunotherapy to limit the activity of antigen presenting cells and T cells in cancer, autoimmune disease, and inflammatory disease^1,2.

EB0126C-2F3-H4/K2 was generated in rabbit for use in immuno-MRM assays using a synthetic peptide derived from human CD40 that is carbamidomethylated at the cysteine, YC[cam]DPNLGLR³. EB0126C-2F3-H4/K2 does not detect CD40 by immunohistochemistry, immunofluorescence, reverse phase protein array, or as recombinant protein by Western blotting⁴. EB0126C-2F3-H4/K2 does detect CD40 by Western blotting in whole cell lysates of buffy coat, HeLa, Jurkat, A549, MCF7, and H226 at the expected molecular weight.

CD40 is expressed by B cells and in primary carcinomas.
