Background
CEACAMs are Ig-related proteins involved in a wide variety of physiological processes, with specific functions dependant on the tissue of expression1. Functions include cell adhesion, pathogen binding, angiogenesis, immune regulation, insulin receptor level regulation, hearing1 as well as cell proliferation, survival, and tumor suppression2. CEACAMs dimerize in homo- and heterophilic interactions with other CEACAMs to perform their functions. CEACAM8 dimerizes with CEACAM1 and CEACAM6 to initiate signaling for various immunomodulatory activities1. When bound to CEACAM1, which is present on other immune cells, CEACAM8 acts as a co-stimulatory factor for B cells or inhibits TLR2 response in lung epithelial cells. When bound to CEACAM6, which is expressed on epithelial cells, neutrophils, and in inflammatory conditions, CEACAM8 blocks CEACAM6 from engaging in other cis or trans interactions with CEACAM1, CEACAM3, and CEACAM5, and thereby restricts signaling via those CEACAMs1. CEACAM8 is attached by GPI anchor to neutrophil surfaces and cannot signal directly. Additionally, CEACAM8 does not bind to known pathogens.

FSAI140R-8C1 was generated in mouse for use in immuno-MRM assays using a synthetic peptide derived from residues 79-98 of human CEACAM8, IIGYVISNQQITPGPAYSNR3. FSAI140R-8C1 detects CEACAM8 by immunohistochemistry, NCI60 protein array, and in Western blots of HeLa and H226 cell lysates4. Western blotting did not detect CEAMCAM8 as recombinant protein or in A549, Jurkat, MCF7, and PBMC whole cell lysates. FSAI140R-8C1 also does not detect CEACAM8 by immunofluorescence.

Antigen Distribution
CEACAM8 is expressed on neutrophils and can be released in a soluble form from intracellular stores. CEACAM8 is also expressed in leukocytes of chronic myeloid leukemia patients.
