CQEEVSHIPAVHPGSFRPK. CPTC Clone ID CPTC-CD74-1 Background CD74 is a transmembrane protein with proinflammatory and immune system functions1. CD74
is the cognate receptor for the proinflammatory cytokine MIF (macrophage migration inhibitory
factor), which modifies the actions of transcription factors and enzymes and also induces
inflammation when secreted. MIF binds to the extracellular domain presented by CD74 to
produce context- and cell-type-specific immune responses. Additionally, the intracellular domain
of CD74, when cleaved, acts as a chaperone for the MHC class II complex or can enter the
nucleus to activate transcription. CD74 regulates transcription in healthy B cells and plays a role
in cancer progression by encouraging cell proliferation, inhibiting cell death, and via
involvement in intercellular communication between regulatory T cells and fibroblasts, epithelial
cells, and B cells in the tumor environment. CD74 is also involved in the etiology of autoimmune
diseases, including multiple sclerosis, rheumatoid arthritis, and autoimmune-associated kidney
disease. CD74 is a target of immunotherapy research2. EB0130A-1G3-H4/K1 was generated in rabbit for use in immuno-MRM assays using a synthetic peptide derived from human CD74 that is carbamidomethylated at the cysteine, C[cam]QEEVSHIPAVHPGSFRPK3. EB0130A-1G3-H4/K1 detects CD74 by immunohistochemistry and by Western blotting as a recombinant protein or in cell lysates of Jurkat and MCF7 cell lines4. EB0130A-1G3-H4/K1 does not detect CD74 by immunofluorescence or reverse phase protein array. Antigen Distribution CD74 is widely expressed in human immune cells, including B cells,
monocytes, activated T cell subsets, and macrophages. CD74 is also expressed by dendritic,
