Due to inherent biochemical properties of antibodies, certain products may be prone to precipitation over time. Precipitation may be removed by aseptic centrifugation and/or filtration. State of Matter Liquid Product Preparation Functional grade preclinical antibodies are manufactured in an animal free facility using only in vitro protein free cell culture techniques and are purified by a multi-step process including the use of protein A or G to assure extremely low levels of endotoxins, leachable protein A or aggregates. Storage and Handling Functional grade preclinical antibodies may be stored sterile as received at 2-8°C for up to one month. DescriptionDescriptionSpecificity Activity is directed against a neuraminidase-sensitive isoform of mouse CD45, specifically CD45RB (also known as B220 or T200). Clone HB220 recognizes a restricted subset of epitopes expressed on the leukocyte common antigen (PTPRC) found in various mouse strains.
Background CD45 is a type I transmembrane protein tyrosine phosphatase (PTP) essential for T and B cell antigen receptor signaling. Different isoforms of CD45, such as CD45RB, are generated through alternative splicing of exons 4, 5, and 6, resulting in extracellular domains with varying glycosylation patterns. CD45RB plays a critical role in modulating the threshold for lymphocyte activation by dephosphorylating members of the Src family kinases. Clone HB220 is highly valued in transplant immunology for its ability to induce long-term allograft survival and immunological tolerance in murine models. Treatment with HB220 has been shown to shift T cell populations from a naive (CD45RB high) state to a memory or regulatory-like (CD45RB low) state, effectively suppressing inflammatory responses in autoimmunity and organ rejection studies. Antigen Distribution CD45RB is expressed on B cells, a subpopulation of T cells (specifically naive T cells), and subsets of natural killer (NK) cells and macrophages.
