Formulation
This recombinant monoclonal antibody is aseptically packaged and formulated in 0.01 M phosphate buffered saline (150 mM NaCl) PBS pH 7.2 - 7.4 with no carrier protein, potassium, calcium or preservatives added. Due to inherent biochemical properties of antibodies, certain products may be prone to precipitation over time. Precipitation may be removed by aseptic centrifugation and/or filtration.
Product Preparation
Recombinant antibodies are manufactured in an animal free facility using only in vitro protein free cell culture techniques and are purified by a multi-step process including the use of protein A or G to assure extremely low levels of endotoxins, leachable protein A or aggregates.
Pathogen Testing
To protect mouse colonies from infection by pathogens and to assure that experimental preclinical data is not affected by such pathogens, all of Leinco’s recombinant biosimilar antibodies are tested and guaranteed to be negative for all pathogens in the IDEXX IMPACT I Mouse Profile.
Storage and Handling
Functional grade preclinical antibodies may be stored sterile as received at 2-8°C for up to one month. For longer term storage, aseptically aliquot in working volumes without diluting and store at ≤ -70°C. Avoid Repeated Freeze Thaw Cycles.

Description

Specificity
Clone MP5-20F3 recognizes an epitope on mouse IL-6.
Background

Background & Biological Significance of Mouse IL-6

Interleukin-6 (IL-6) is a highly pleiotropic, 21–28 kDa cytokine that functions as an essential mediator of host defense, acute phase responses, hematopoiesis, and immune regulation. Secreted by a variety of cell types—including monocytes, T cells, fibroblasts, and endothelial cells—mouse IL-6 can behave as both a pro-inflammatory cytokine and an anti-inflammatory myokine.

Upon binding to the cell-surface IL-6 Receptor (IL-6R), it initiates a complex with gp130, triggering the downstream JAK/STAT3 signaling pathway. Dysregulated or chronically elevated levels of mouse IL-6 are closely linked to autoimmune disorders (such as systemic rheumatoid arthritis), cytokine release syndrome (CRS), and the immunosuppressive restructuring of the tumor microenvironment (TME). Consequently, blocking the IL-6 pathway is a primary focus in preclinical oncology and inflammation research.

Mechanism and Applications of Anti-Mouse IL-6 Clone MP5-20F3

The MP5-20F3 monoclonal antibody specifically targets and binds to mouse interleukin-6, acting as a potent neutralizing antibody. By sterically hindering the interaction between mouse IL-6 and its receptor, clone MP5-20F3 completely blocks gp130-mediated signal transduction and downstream STAT3 activation.

In preclinical rodent models and functional assays, clone MP5-20F3 is widely utilized for:
* **In Vivo Cytokine Neutralization & Depletion: Investigating the functional impact of IL-6 systemically or locally.
* **Tumor Microenvironment Reshaping: Evaluating T-cell infiltration, Treg modulation, and malignant pleural effusion (MPE).
* **In Vitro Functional Assays: Serving as a reliable capture antibody for sandwich ELISA development or ELISPOT applications.

The Recombinant Advantage for Preclinical In Vivo Studies

Unlike traditional hybridoma-derived antibodies that may suffer from cell-line drift or animal-protein contamination, Leinco’s recombinant anti-mouse IL-6 antibody (Clone MP5-20F3) is engineered using defined genetic sequences and expressed in an animal-free HEK-293 mammalian expression system. This ensures exceptional batch-to-batch consistency and high-purity monomeric conformation.

Formulated specifically as an in vivo Functional Grade GOLD reagent, this antibody is aseptically packaged in PBS (pH 7.2–7.4) without any preservatives, carrier proteins, potassium, or calcium. This ultra-clean formulation, combined with guaranteed low endotoxin levels (≤ 1.0 EU/mg), prevents non-specific immune activation, making it the premier choice for highly sensitive animal models and preclinical therapeutic validation.
