Immunofluorescence: use at 0.2-2ug/ml. These are recommended concentrations. User should determine optimal concentrations for their application. Positive control: S. cerevisiae lysate. Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change. DescriptionSpecificity This antibody recognizes a ~70 kDa protein corresponding to the predicted molecular mass of HDEL in Drosophila, yeast, and various plants (barnyard grass, beet, cotton, mung bean, sorghum, and wheat). Background The Hsp70 family includes four highly conserved proteins (Hsp70, Hsc70, Grp75, and Grp78) which function as molecular chaperones, in translocation of polypeptides across cell membranes and to cell nuclei, and in proper folding of nascent polypeptide chains. Sorting of proteins by the Hsp70 family relies on a C-terminal tetrapeptide signal, usually KDEL in animal cells, and HDEL in S. cerevisia.
