Formulation
This monoclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 - 7.4 containing 5.0% w/v trehalose with no calcium, magnesium or preservatives present.
Storage and Handling
The lyophilized antibody can be stored desiccated at -20°C to -70°C for up to twelve months. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid repeated freeze thaw cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Human BMP-7 this monoclonal antibody can be used at a concentration of 1-2 µg/ml. This monoclonal antibody should be used in conjunction with compatible second-step reagents such as PN:M114 and a chromogenic substrate such as PN:T343. The detection limit for Human BMP-7 is 25 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is PN:B525.
Additional Applications Reported In Literature ?
IHC (NBF/Par.): This antibody should give satisfactory staining results when used at a concentration of 25 µg/ml. The recommended secondary antibody for IHC is PN:M114. For chromogenic detection with high signal and low background use PN:D100 or PN:K107.
Neutralization: This antibody is useful for neutralization of Human BMP-7 bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Human BMP-7 (at 1.0 ng/ml) is 1.5 - 6 µg/ml.

Specificity
Mouse Anti-Human Bone Morphogenetic Protein 7 (BMP-7) (Clone 164311) recognizes an epitope on Human BMP-7. This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype. This antibody showed approximately 25% cross-reactivity with rhBMP-6 and no cross-reactivity with rhBMP-2, -3, -4, -5 or -8.
Background
TGF-β family members are key modulators of cell proliferation, differentiation, matrix synthesis, and apoptosis. As implied by their name, BMPs initiate, promote, and regulate the development, growth and remodeling of bone and cartilage. In addition to this role, BMPs are also involved in prenatal development and postnatal growth, remodeling and maintenance of a variety of other tissues and organs. BMP-7, also known as osteogenic protein-1 or OP-1, is a potent bone inducing agent, which in the presence of appropriate osteoconductive carrier (e.g. collagen sponge or synthetic hydroxyapatite) can be used in the treatment of bone defects. A bone-graft substitute, called OP-1TM implant, made of recombinant human BMP-7 associated with bovine bone-derived collagen, has recently been approved by the FDA as a device for treating critical-size bone fractures. The potential use of BMP-7 in dental reconstructive surgeries is currently under investigation. Recombinant human BMP-7 is a 28.8 kDa homodimeric glycoprotein consisting of two 116 amino acid subunits, which correspond to amino acid residues 316 to 431 of the full-length BMP-7 precursor.

BMP's are evolutionarily conseved acrross the animal species. For example human and mouse share 98% sequence identity of the mature protein. BMP-7 is expressed as a large precursor protein cleaved at the dibasic (RXXR) site to release the carboxy-terminal domain. Biologically active BMP-7 has been shown to exhib its cellular responses by the formation of hetero-oligomeric complexes of type I and II serine/threonine kinase receptors. BMP's 5,6,7,and 8 are in the same subgroup based on amino acid sequence similarity.
