The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Human IL-17E this polyclonal antibody can be used at a concentration of 0.1 - 0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as PN:G505 and a chromogenic substrate such as PN:T343. The detection limit for Human IL-17E is 2 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is PN:I-199.

Neutralization: This antibody is useful for neutralization of IL‑17E/IL‑25-induced CXCL1/GRO alpha secretion in the HT‑29 human colon adenocarcinoma cell line. The antibody dose required to neutralize 50% (ND50) of the biological activity of Human IL-22 (at 1 ng/ml) is 0.5 - 2.5 µg/ml.

Background
Interleukin (IL)-17E, is a novel member of the IL-17 family of cytokines1 and a unique pleiotropic cytokine and may be an important mediator of inflammatory and immune responses.2 IL17B is also a ligand for the cytokine receptor IL17BR. IL-17E induces activation of NF-kappaB and stimulates production of the proinflammatory chemokine IL-8. 1 IL-17E may contribute to the induction and maintenance of eosinophilic inflammation in the airways by acting on lung fibroblasts and therefore has a role in asthma pathophysiology.3
