Validation Notes Clone M3/84 has been validated for use on PhenoCycler® using [Mouse/Human] [Tissue] [Tissue Type either FFPE or FF] tissue. Antibody and Reporter DetailsReactivity Species Mouse Host Species Rat Concentration 0.5 mg/ml Immunogen Surface glycoproteins from macrophage membranes of peritoneal exudate cells of C57BL/6 mice Formulation This purified antibody is formulated in 0.01 M phosphate buffered saline (150 mM NaCl) PBS pH 7.4. Storage and Handling This antibody is stable for at least one week when stored at 2-8°C. For long term storage, aliquot in working volumes without diluting and store at -20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. Applications and Recommended Usage? Quality Tested by Leinco CODEX® This M3/84 antibody is formulated to simplify the antibody preparation needed when performing a PhenoCycler® barcode conjugate. The suggested concentration is 1.0 mg/ml. Other Applications Reported In Literature ? FC, ICC, IHC, IP, RIA Country of Origin USA Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change. DescriptionSpecificity M3/84 activity is directed against mouse CD107b (Mac-3), also known as LAMP-2. Antigen Distribution CD107b (LAMP-2) is a constituent of the lysosomal membrane. CD107b
is present in macrophages, granulocytes, peripheral blood lymphocytes, bronchial epithelium,
alveolar lining cells, intestinal epithelium, and some dendritic and endothelium cells. Background Macrophages function in phagocytosis of foreign organisms and materials as well as resistance to
tumors1. Additionally, they play roles in T lymphocyte immune responsiveness, inflammatory
responses, and regulation of hematopoiesis. Lysosomes are the membrane-bound vacuoles that
digest foreign materials and perform specialized autolytic functions2. Lysosomes also play a role
in antigen presentation, cholesterol metabolism, and hydrolysis of exogenous materials3 . CD107b (LAMP-2, Mac-3) is a member of the lysosome-associated membrane protein (LAMP) family and is an important component of the lysosomal membrane involved in autophagy3,4 . CD107b is a highly glycosylated transmembrane protein, with glycosylation being necessary for its stability5,6. Mice deficient in CD107b have a severe phenotype, with 50% dying aged 20-40 days6. In humans, mutations in CD107b lead to Danon disease. The immunogen used to generate M3/84 was produced by first purifying surface glycoproteins from macrophage membranes of peritoneal exudate cells of C57BL/6 mice5. Next, previously recognized antigens were removed by immunoadsorbent columns to obtain a glycoprotein- enriched fraction. Then, (Lewis x BN) F1 rats were injected and splenocytes fused to mouse myeloma NS1 for hybridoma production.
