This biotinylated antigen affinity purified polyclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 containing 50 µg of bovine serum albumin per µg of antibody with no calcium, magnesium, or preservatives present.

Storage and Handling
The lyophilized, biotinylated antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for up to twelve months from date of receipt. The reconstituted biotin conjugate can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted conjugate, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Mouse IL-21 this biotin conjugate can be used at a concentration of 0.1 - 0.2 µg/ml. This biotin conjugate should be used in conjunction with compatible second-step reagents such as PN:A106 and a chromogenic substrate such as PN:T343. The detection limit for Mouse IL-21 is 1 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.
ELISA Detection: Suitable for use at concentration of 0.1-0.4 µg/mL. Use in combination with: Capture Reagent: Mouse IL‑21 Antibody. Standard: Recombinant Mouse IL-21 Protein.

Background
Interleukin 21 (IL-21) is a T cell-derived pleiotropic cytokine that has potent regulatory effects on cells of the immune system, including natural killer (NK) cells and cytotoxic T cells that can destroy virally infected or cancerous cells.1,2 This cytokine induces cell division/proliferation in its target cells. IL-21 elicits its effects on immune cells by interacting with a cell surface receptor known as the interleukin 21 receptor, IL-21R, which is expressed in bone marrow cells and various lymphocytes. IL-21 is closely related to IL-2 and IL-153 and is a key factor in the transition between innate and adaptive immune responses.4 The critical role of IL21 in promoting humoral immune responses makes it an important focus of potential therapeutic interventions in conditions characterised by overproduction of pathogenic autoantibodies.5
