Description

Background
As with other Horseradish Peroxidase substrates, ABTS reactivity can be improved with the addition of a chemical enhancer. ABTS 1 Component HRPO Microwell Enhancer is optimized for use with ABTS HRPO Microwell Substrate One Component "Ready Use" (Leinco Prod. No.: A202). When ABTS 1 Component HRPO Microwell Enhancer is used, ABTS reactivity can be increased from 50% to 150%. See the graph below for an example of the resulting increase in reactivity. This could be useful for improving assay sensitivity or to help conserve assay components. ABTS 1 Component HRPO Microwell Enhancer is not recommended for membrane or immunohistochemical applications that require a precipitation reaction product.
Directions for Use
ABTS 1 Component HRPO Microwell Enhancer is supplied as an 11X concentrate. For microwell applications, add 1 mL of ABTS 1 Component HRPO Microwell Enhancer to 10 mL of ABTS HRPO Microwell Substrate One Component "Ready Use" (Leinco Prod. No.: A202). Mix gently for approximately 1 minute.

Add 100 μl of the enhanced substrate solution to each well of the 96 well ELISA plate. Once a soluble blue-green reaction product develops, the plate can be read at 405 nm to 410 nm. The absorbance values of the sample should be monitored so that a linear curve can be plotted. The substrate reaction can be stopped by addition of 100 μl of a stop solution such as 1% sodium dodecyl sulfate (SDS) or 405/410 nm Stop Reagent for ABTS Microwell (Leinco Prod. No.: A204). OD values should be read at 405 nm to 410 nm with or without the addition of stop solution. Estimated incubation times for substrate range from 15 to 30 minutes.
