Specificity
1B4 activity is directed against mouse TIGIT.
Background
TIGIT is an immune checkpoint receptor that inhibits critical immune responses, including T cell priming by dendritic cells and the tumor-killing activity of NK and cytotoxic T cells. Because it also enhances the suppressive function of regulatory T cells, TIGIT has emerged as a significant target for cancer immunotherapy, often in combination with the PD-1/PD-L1 pathway. By conjugating the 1B4 monoclonal antibody to the CF®647 fluorophore, researchers can effectively label TIGIT-expressing cells for ex vivo analysis. This combination is particularly beneficial for high-sensitivity flow cytometry, as the bright, photostable signal of CF®647 allows for the precise identification and functional assessment of TIGIT-positive populations—such as activated and memory T cells—within complex biological samples.
