Immunogen
Purified Recombinant Rat IL-1β (>98%)
Endotoxin Level
<0.01 EU/µg as determined by the LAL method
Formulation
This antigen affinity purified polyclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 containing 5.0% w/v trehalose with no calcium, magnesium, or preservatives present.
Storage and Handling
The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Applications and Recommended Usage?
Quality Tested by Leinco
ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 0.2-0.8 µg/ml. A suitable detection antibody is PN:I-248 at a concentration of approximately 0.1-0.4 µg/ml.
Western Blotting: To detect Rat IL-1β this polyclonal antibody can be used at a concentration of 0.1-0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as PN:G505 and a chromogenic substrate such as PN:T343. The detection limit for Rat IL-1β is 5 and 2 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.
Other Applications Reported In Literature ?
Immunocytochemistry: Suitable at concentration of 5-15 µg/mL.
Neutralization: Measured by its ability to neutralize IL‑1 beta /IL‑1F2-induced proliferation in the D10.G4.1 mouse helper T cell line. Symons, J.A. et al. (1987) in Lymphokines and Interferons, a Practical Approach. Clemens, M.J. et al. (eds): IRL Press. 272. The Neutralization Dose (ND50) is typically 2-10 µg/mL in the presence of 10 ng/mL Recombinant Rat IL‑1 beta /IL‑1F2.
Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change.

Description

Specificity
Rabbit Anti-Rat Interleukin-1β (IL-1β) recognizes Rat IL-1β. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.
Background
IL-1R1 is an 80 kD transmembrane glycoprotein and a member of the Ig superfamily. IL-1 binds both IL-1R1 and IL-1R2. The ligands IL-1α and IL-1β only commence when binding IL-1R1 because IL-1R2 acts as a decoy receptor and competitively antagonizes the binding of IL-1α and IL-1β, thereby reducing their biological effects. Binding of the ligands to IL-1R1 is enhanced by an accessory protein, IL-1R-AcP. IL-1α can exist as either a 31 kD precursor or an 18 kD mature form. IL-1α is a proinflammatory cell-associated cytokine and IL-1β is a 17.5 kD proinflammatory secreted cytokine. Both IL-1α and IL-1β bind to the same receptor and have similar properties that include stimulation of thymocyte proliferation via IL-2 release, B-cell maturation and proliferation, mitogenic FGF-like activity and the ability to stimulate the release of prostaglandin and collagenase from synovial cells. Furthermore, IL-1β mediates a variety of immune and inflammatory responses. IL-1β is a major cause of severe inflammation in the mouse model of Crohn’s Disease, which makes IL-1β an important target for this disease.
