Purified Recombinant Rat IL-4 (>98%)

This antigen affinity purified polyclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 containing 5.0% w/v trehalose with no calcium, magnesium, or preservatives present.

The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Neutralization: Measured by its ability to neutralize IL‑4-induced proliferation in the rat splenocytes. The Neutralization Dose (ND₅₀) is typically 0.05-015 µg/mL in the presence of 4 ng/mL Recombinant Rat IL‑4 and 10 µg/mL PHA.

Rabbit Anti-Rat Interleukin-4 (IL-4) recognizes Rat IL-4. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. IL-4 is a 15-19 kD pleiotropic immune cytokine secreted by activated TH2 cells. It has many biological roles, including the regulation in humoral and adaptive immunity, the inhibition of bone resorption, the stimulation of activated B-cell and T-cell proliferation, and initiation of allergic responses. Moreover, IL-4 induces a positive feedback loop by stimulating differentiation of naive helper T cells, or Th0 cells, to Th2 cells which in turn secrete more IL-4. Increased IL-4 production and overproduction of IL-4R are found in many cancers. IL-4 has a significant effect on tumor progression by increasing apoptosis resistance via tumor growth enhancement. IL-4 may also play a part in the immune dysregulation that occurs in most HIV-1 infected patients.
