Lyophilized
Storage and Handling
The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Applications and Recommended Usage?
Quality Tested by Leinco
Western Blotting: To detect Rat IL-5 this polyclonal antibody can be used at a concentration of 0.1-0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as PN:G505 and a chromogenic substrate such as PN:T343. The detection limit for Rat IL-5 is 20 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is PN:I-534.
Additional Applications Reported In Literature ?
Immunohistochemistry on Frozen Sections: This antibody should give satisfactory staining results when used on frozen sections at a concentration of 15 µg/ml.
Neutralization: This antibody is useful for neutralization of Rat IL-5 bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Rat IL-5 (at 1.25 ng/ml) is 2-6 µg/ml.
Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change.

Description

Specificity
Goat Anti-Rat Interleukin 5 (IL-5) recognizes Rat IL-5. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.
Background
IL-5 is a 26 kD TH2 cytokine and a homodimeric glycoprotein that is part of the hematopoietic family. IL-5 stimulates B cell growth as well as increases Ig secretion. It is the main factor that promotes the terminal differentiation of eosinophil progenitors and enhances the effector capacity of mature eosinophils. IL-5 is a major regulator of eosinophil accumulation in tissues. In fact, in Hodgkin lymphoma, the characteristically observed eosinophilia is suspected to result from an increased production of IL-5. IL-5 may also be the cause of several allergic diseases including asthma, where it is thought that eosinophils have a significant role in the disease pathology. There are several mAbs currently in use that target IL-5 for the treatment of severe eosinophilic asthma.
