This antigen affinity purified polyclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.4 containing 5.0% w/v trehalose with no calcium, magnesium, or preservatives present.
Storage and Handling
The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Mouse CXCL10 this polyclonal antibody can be used at a concentration of 1 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents and a chromogenic substrate such as PN:T343.

Immunohistochemistry on Frozen Sections: This antibody should give satisfactory staining results when used on frozen sections at a concentration of 5-15 µg/ml.
Neutralization: This antibody is useful for neutralization of Mouse CXCL10 bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Mouse CXCL10 (at 0.5 ug/ml) is 5-25 µg/ml.

Background
Chemokine (C-X-C motif) ligand 10 (CXCL10) or IP-10 is a small cytokine belonging to the CXC chemokine family. CXCL10 is secreted by several cell types in response to IFN-γ. These cell types include monocytes, endothelial cells and fibroblasts.1 CXCL10 has been attributed to several roles, such as chemoattraction for monocytes/macrophages, T cells, NK cells, and dendritic cells, promotion of T cell adhesion to endothelial cells, antitumor activity, and inhibition of bone marrow colony formation and angiogenesis.2,3 CXCL10 plays a role in the regulation of tubular cell proliferation following renal ischemia-reperfusion injury.4
