This secondary antibody is lyophilized from 0.01M phosphate buffered saline (PBS) pH 7.6 containing 0.25M NaCl, 15 mg/ml IgG free BSA and 0.05% sodium azide as a preservative.
Storage and Handling
This lyophilized secondary antibody conjugate should be stored desiccated at 2-8°C. The reconstituted AMCA conjugate containing glycerol should be stored at -20°C in a manual defrost freezer.

Indirect ELISA: This antibody can be used in an indirect detection ELISA at 0.5-1.0 µg/ml with a suitable second step reagent such as PN:G505. The detection sensitivity of this indirect ELISA for Human Leptin is approximately 0.08 ng/well.

IHC (NBF/Par.): This antibody may be used to stain nuclear membranes in cells, tissue preparations or subcellular fractions. The antibody works well with cells and tissue sections fixed with 2% paraformaldehyde and requires no dilution. Tonsil is the recommended positive control tissue.

Neutralization: The peptide may be used to neutralize the binding activity of PN:Z100 by incubation of an equal volume of the peptide and antibody for 30 minutes at 37°C for Western blotting experiments.

Western Blotting: To detect Human CASP8 by Western blot analysis, this antibody can be used at a concentration of 1.0-2.0 µg/ml (1:100 Dilution). When used in conjunction with compatible second step reagents such as PN:M114 and a chromogenic substrate such as PN:T343, the detection limit for Human CASP8 is 1.0 ng/lane under either reducing or non-reducing conditions.
Specificity
Goat Anti-Rat IgG (H&L) recognizes Rat IgG heavy and light chains. This secondary antibody was purified using antigen affinity chromatography and adsorbed against serum proteins and immunoglobulins to minimize species cross-reactivity.
